Adhesion Class GPCRs in GtoPdb v.2023.1

چکیده

Adhesion GPCRs are structurally identified on the basis of a large extracellular region, similar to Class B GPCR, but which is linked 7TM region by GPCR autoproteolysis-inducing (GAIN) domain [10] containing proteolysis site (GPS). The N-terminal often shares structural homology with adhesive domains (e.g. cadherins, immunolobulin, lectins) facilitating inter- and matricellular interactions leading term adhesion [104, 418]. Several receptors have been suggested function as mechanosensors [320, 288, 396, 38]. Cryo-EM structures 7-transmembrane several determined recently [292, 21, 403, 212, 300, 302, 431, 293]. nomenclature these was revised in 2015 recommended NC-IUPHAR Consortium [125].

Adhesion GPCRs in Kidney Development and Disease

Chronic kidney disease (CKD) represents the fastest growing pathology worldwide with a prevalence of >10% in many countries. In addition, kidney cancer represents 5% of all new diagnosed cancers. As currently no effective therapies exist to restore kidney function after CKD- as well as cancer-induced renal damage, it is important to elucidate new regulators of kidney development and disease as ...

Versatile Signaling Activity of Adhesion GPCRs.

The adhesion G protein-coupled receptors (aGPCRs) are a family of 33 receptors in humans that are widely expressed in various tissues and involved in many diverse biological processes. These receptors possess extremely large N-termini (NT) containing a variety of adhesion domains. A distinguishing feature of these receptors is the presence within the NT of a highly conserved GPCR autoproteolysi...

The Origin of GPCRs: Identification of Mammalian like Rhodopsin, Adhesion, Glutamate and Frizzled GPCRs in Fungi

G protein-coupled receptors (GPCRs) in humans are classified into the five main families named Glutamate, Rhodopsin, Adhesion, Frizzled and Secretin according to the GRAFS classification. Previous results show that these mammalian GRAFS families are well represented in the Metazoan lineages, but they have not been shown to be present in Fungi. Here, we systematically mined 79 fungal genomes and...

Class B GPCRs: a hidden agonist within?

Class B G protein-coupled receptors (GPCRs) regulate a wide range of endocrine and neuroendocrine functions and are endogenously stimulated by moderately large peptide hormones. Current evidence suggests that the carboxyl termini of cognate peptides bind to the amino terminus of their G protein-coupled receptors (GPCRs) and that the peptides' amino terminal segments then dock to the heptahelica...

Super-complexes of adhesion GPCRs and neural guidance receptors.

Latrophilin adhesion-GPCRs (Lphn1-3 or ADGRL1-3) and Unc5 cell guidance receptors (Unc5A-D) interact with FLRT proteins (FLRT1-3), thereby promoting cell adhesion and repulsion, respectively. How the three proteins interact and function simultaneously is poorly understood. We show that Unc5D interacts with FLRT2 in cis, controlling cell adhesion in response to externally presented Lphn3. The ec...
